Abstract:
Original Grifola frondosa strain GF3 isolated from Slovenian forests was cultivated in a liquid substrate based
on glucose, with polypeptone as the source of nitrogen and mineral salts, and on a solid substrate based on
milled whole maize plant (Zea mays), olive oil and mineral salts.
Production of extracellular and intracellular
polysaccharides in both substrates was studied.
In submerged cultivation, in 28 days 3.65 mg/L of extracellular
and 1.30 mg/L intracellular polysaccharide were produced.
In solid-state cultivation, in 38 days 3.80 mg/g
of extracellular and 0.70 mg/g of intracellular polysaccharide were produced.
Polysaccharides were further
separated by ion-exchange, and gel and affinity chromatography.
The isolated polysaccharides were mainly
ß-D-glucanes.
