Abstract: A new heteropolysaccharide, P60W-1, isolated from a medicinal fungus, Phellinus igniarius, has a molecular weight of 1.71 × 104 Da and is composed of fucose, glucose, mannose, galactose and 3-O-Me-galactose in the ratio 1:1:1:2:1. Structure investigation of the P60W-1 was carried out using sugar and methylation analysis, together with 1H and 13C NMR spectroscopy, including COSY, TOCSY, NOESY, HSQC and HMBC experiments. The repeating unit of the polysaccharide is established as the following structure. The result also showed that P60W-1 has an in vivo antitumour effect at the most effective dose of 100 mg/kg, inhibiting the growth of liver tumor H22 and Lewis lung carcinoma (LLC) tumour in mice by 48% and 37%, respectively.
