Abstract: Volvariella volvacea, commonly known as the Chinese straw mushroom, is an important edible fungus of the tropics and subtropics. Chilling injury has been found in this mushroom. Low temperatures induce cellprotein degradation and result in mycelium autolysis in V. volvacea, creating problems for cultivation and fruiting-body storage. In this study, protoplast of V. volvacea was prepared for complex mutagenesis with UV, 60Co-γ and diethyl sulfonate. Fixing low temperature as the selection index, cold-resistant strain VH3 was obtained from 1500 mutants. It could survival about 10 days at 4°C. Isozyme analysis showed that variation existed between VH3 and a control strain in terms of both zymogram pattern and activity of peroxidase and esterase isoenzymes. The mean mutation coefficient was 0.213 by random amplified polymorphic DNA analysis. The biological efficiency of VH3 at 26, 28 and 30°C was higher than that of the control strain. The difference was significant. The optimal cultivation temperature of VH3 was 28°C, but it can also be cultivated at 26°C. The strain was genetically stable, and could be cultivated at comparatively low temperatures.
