Abstract: We used the ‘fruiting body’ Agrobacterium-mediated genetic transformation method developed at Penn State to express several novel genes in the button mushroom, Agaricus bisporus. Coupled with strain J1901, a proprietary sporeless variety developed at Sylvan, a contained system using a mushroom biomass platform was created that allows for the development of new bioengineered products. Herein, we summarize the methodology used and findings obtained for four different transgenes. Verification of integration and expression of the transgenes included analyses by PCR, Southern blotting and antibody and functional enzyme assays. This work demonstrated the potential of this technology for utilizing mushroom biomass for unique agricultural, food and pharmaceutical applications.
