Abstract: Reverse transcription-random amplified polymorphic DNA analysis was used to isolate several thermotolerance-related DNA fragments in Agaricus bisporus. One fragment was cloned and sequenced and corresponded to a portion of the tRNAVal gene. Using a differential display approach, several other thermotolerance-related DNA fragments were cloned and sequenced. The results indicated that one fragment was an unreported gene, a second fragment shared high homology in amino acid sequence with the partial karyopherin docking complex of the nuclear pore complex gene in Saccharomyces cerevisiae and a third fragment had amino acid sequence homology with the vitamin-K-dependent protein S.
