Abstract: We were interested in identifying genes associated with differentiation and development in Agaricus bisporus. Upregulated genes identified during the rapid expansion phase of the sporophore included sugar transporter gene Ab sut1, putative riboflavin-aldehyde-forming-enzyme gene and three novel morphogenes. Further, glucose/hexose transporter gene (Ab sut2), glucose regulated protein gene, ABC transporter gene, chitin synthase gene, and lectin gene (abl1) were PCR cloned from A. bisporus. Results of Northern analyses indicated an upregulation during the early differentiation phase. During mushroom development, Ab sut1 transcripts increased more than 100-fold. This gene showed a 33% identity with other sugar transporters, but differed in its substrate preference. Interestingly, Ab sut2 showed strong homology to fungal glucose/hexose transporters and its homologs appear to be present in A. bitorquis and Coprinus cinereus. Abl1 showed interesting features in its genomic organization and expression patterns.
