Title: Maturation and activation of latent tyrosinase from Agaricus bisporus

Abstract:

Latent mushroom (Agaricus bisporus) tyrosinase was activated by a number of methods that result in either conformational changes in the enzyme, or to structural (proteolytic) alterations. Activated enzymes were analyzed with denaturing SDS/PAGE and Western blotting. Kinetic features of both activation process and final active enzymes were characterized. Based on these in vitro observations, a maturation and activation model for mushroom tyrosinase was postulated, that may account for senescence, damage and disease related browning of mushrooms.
