Abstract: We have initiated a comprehensive survey of bacterial and fungal microorganisms in mushroom compost using the sequence of genes encoding small subunit ribosomal RNA (rDNA) as a molecular tag. Microorganisms in Phase II compost were mechanically lysed and their DNA was purified. The DNA was subjected to polymerase chain reaction using pairs of primers designed for amplifying segments of 16S or 18S rDNA. Libraries of rDNA were constructed using the amplified rDNA, and clones in the libraries were sequenced. Taxonomie classification of these sequences was performed by comparison with complete or partial rDNA sequences of previously identified microorganisms stored in public databases using the BLAST algorithm. Initial results indicate that commercial mushroom subsfrate supports many unreported microorganisms that represent over 7 different divisions of bacteria and 4 different orders of fiingi.
