Abstract: The domains V4, V6 and V9 of the mitochondrial small subunits (SSU) rDNAs have been successfully used as species-specific markers for the taxonomie characterization and for phylogenetic studies within the Agrocybe and Pleurotus genera. Using PCR amplifications and sequencing, these smdies were extended to species belonging to the Agaricaceae, Coprinaceae and Tricholomataceae families. Lengths and sequences were compared among the V4, V6 and V9 domains, the individual frequencies of positions with substitutions and of insertion/deletions events were calculated. These frequencies vary according to the taxonomie group studied. In each group of species, conserved motifs generally located at the 5' and/or 3' ends of the three domains were evidenced, these consensus sequences seems to be specific of he taxonomie groups. Comparisons of the V4 domains among Agrocybe aegerita related strains suggested an evolutionary pathway involving inverted repeated sequences and a two-step insertion event. The phylogenetic tree constructed from the sequences of variable mitochondrial domains was congruent with phylogenies based on morphological, nuclear and mitochondrial characters.
