Abstract: The complete sequence of the Agrocybe aegerita mitochondrial nd6 gene was achieved from a Sca I - Ava I mitochondrial fragment. The nd6 gene is located in the upstream region of the LSU-rRNA gene, it encodes for a protein of 215 aa rich in leucine and isoleucine residues. Sequence comparison with ND6 proteins from plants, animals and fungi showed that beside the variability of the NH2 and COOH termini of the protein, three regions (RI, R2 and R3) were more conserved, and could be involved in the activity of the ND6 protein. A phylogenetic analysis based upon ND6 sequences suggests that, in contrast with nuclear phylogeny reports, mitochondrial fungal sequences are more closely related to plant and algae sequences than to animal sequences, confirming recent studies based upon mitochondrial genes. This is the first report of the complete nucleotide sequence of a mitochondrial nd6 gene of any member of the Basidiomycota division.
