Abstract: Pleurotus cultivation accounts for a quarter of the total world mushroom production To obtain a rapid and reliable method for identification, RAPD experiments were earned out to take specific genetic markers of Pleurotus ostreatus isolates Genomic DNA from various isolates of P ostreatus was subjected to RAPD amplification using random 10 mers and 20 mers primers One of the twelve primers used yielded a fragment of about 600bp present in all isolates of P ostreatus This fragment was cloned and used as hybridization probe for species and strain identification This DNA probe, pBP20, was hybridized to membranes displaying the respective RAPD products of Pleurotus species and of P ostreatus This clone was used to probe for restriction fragment length polymorphisms (RFLPs), and the probe was sequenced Analysis of data showed that P ostreatus was distinct from other Pleurotus species even though P florida and P pulmonarius have been confused in taxonomical criteria The DNA probe described here is very specific and reliable for the identification of Pleurotus species
