Title: Use of a SCAR marker for cap color in Agaricus bisporus breeding programs

Abstract:

L43 is a marker that is closely linked to the locus for cap color (PPC1). This marker was originally a RAPD band which co-segregated with cap color in a cross between a Horst U-l ® homokaryon (U1A) and a homokaryon derived from a strain collected in Alberta, Canada (1551B). When we tested the RAPD locus on a panel of different genotypes we noted that the band was present in some types and absent in others. Consequently the marker was useful in some but not all crosses. In order to make the marker more useful the band was cloned and sequenced from each end. Using the sequence data we designed a pair of PCR primers that amplified a band of the same size in all test strains. In order to resolve polymorphism the PCR product was digested with the restriction enzyme Cfo I. Using this marker we were able to select for mushroom cap color without a requirement for crossing and fruiting.
