Title: Enzymology and molecular biology of Agaricus bisporus tyrosinase

Abstract:

One full length cDNA fragment of 1.9 kB (AbPPOl) and a partial one of 1.4 kB (AbPP02) were isolated from a cDNA-library of Agaricus bisporus Ul. The open reading frame of AbPPOl encodes a protein of 63.9 kD with significant homology to Neurospora crassa protyrosinase, whereas no homology to A. bisporus laccase was detected. Southern blot analysis using AbPPOl as a probe, indicated one chromosomal copy number for AbPPOl and a comparison with other Agaricus strains and species revealed a polymorphic gene organization. Expression of mRNA sequences that are recognized by partial clones related to AbPPOl is speciesdependent. Also the occurrence of specific tyrosinase iso-forms varied between isolates and species; as in some cases mRNA's are being expressed but no active enzyme can be detected, post-transcriptional regulation is likely.
