Title: Cryo SEM study of changes in tissue anatomy of harvested sporophores of Agaricus bisporus

Abstract:

Cryo-fracture is a newly-applied preparation technique to observe mushrooms under the Scanning Electron Microscope. The technique has revealed anatomical details of sporophore tissues without the problem of exudate smearing and cell displacement. During postharvest senescence a proportion of cells undergo total collapse while the remaining cells remain fully turgid. The number of cells autolysing in this way increases during the storage period leaving a network of intact cells and greatly enlarged intercellular spaces. Mushroom senescence appears to be localised and controlled at the level of individual cells.
