Abstract: DNA based techniques, including RFLP and RAPD analyses, have been used to identify isolates of Mycogone perniciosa, Trichoderma harzianum and Verticilliumfungicola var. fungicola isolates from Agaricus bisporus or mushroom compost. Aggressive isolates of T. harzianum can be distinguished from morphologically similar non-aggressive isolates. A high level of genetic similarity was apparent within the aggressive isolate group which may indicate that outbreaks within the British Isles have originated from a single source. M. perniciosa could be separated from M. rosae, and analysis of V. fungicola isolates revealed distinct genetic pools that appear to be geographically isolated. The basic methodology of each technique is described.
