Title: Rapid extraction of a bacteria free fraction inducing blotch symptoms on Agaricus bisporus

Abstract:

The principle responsible for brown blotch on mushrooms was isolated by Peng in 1986; it is a toxin, tolaasin, secreted in the growth medium by Pseudomonas tolaasii. This toxin was purified by Nutkins et al. (1991) but with a low yield. We have improved the purification method by the use of a solvent and by the way P. tolaasii was cultivated. The aim of the project is to use the toxin as a tool in a biological test for breeding work. The purification was improved by mixing the bacteria, cultivated on agar medium, with butanol. The toxin was then tested on mushrooms of different susceptibility. The use of the partially purified toxin to screen for resistance is now preferred to the use of living bacteria.
