Abstract: After Yun Zhi mycelia or fruitbodies were extracted with hot water and precipitated with alcohol, the Yun Zhi mycelial (MWA) or Yun Zhi fruit body extract (FWA) was obtained. Then they were respectively treated with anthrone and an analysis was made by means of ultraviolet/visible light spectra. The result shows that the maximum absorption wavelengths of MWA and FWA are 420 and 671 nm respectively. After the water solution of MWA is treated with ninhydrin, we see that its maximum absorption wavelength is 586 nm; but there is no obvious absorption of FWA. Through a comparison of the infrared spectra of MWA and FWA, we discover that there is an obvious absorption of MWA at 1380 cm"1, while there is no obvious absorption of FWA at that position. By phenol sulphuric acid and Lowry methods to determine the contents of polysaccharide and protein, we find that the glucose contents of MWA and FWA are 36% and 48% respectively and the protein contents are 33% and 21% respectively. With a rotatory instrument (Polarimeter) to measure the specific rotatory power of MWA and FWA, we find that of MWA is -0.04 and that of FWA is -0.69. With DEAE-cellulose column chromatography and gas chromatography analysis, we find that the main ingredient of MWA (MWA-C) contains 5 kinds of monosaccharides: glucose, mannose, xylose, rhamnose and arabinose; while the main ingredient of FWA (FWA-A) contains 2 kinds of monosaccharides: glucose and rhamnose. The FWA-A is not adsorbed on DEAE-cellulose. Its molecular weight is 6,500 Da. The MWA-C is adsorbed on DEAE-cellulose and its molecular weight is 26,000 Da.
