Abstract: After the fermentation cultivation of mycelia of Lentinus edodes for 72 hours, they were extracted with hot water and precipitated with alcohol. Then the water extract was isolated by DEAE-cellulose column chromatography twice and purified by Sephadex G-100 column chromatography. After that we obtain two kinds of polysaccharides: Le- 2-1 and Le-2-2. Through the identification by means of HPLC and electrophoresis with acetate-cellulose membrane, both Le-2-1 and Le-2-2 are of homogeneous composition. The purity of Le-2-1 is 99.991% and Le-2-2 is 100%. Their compositions of monosaccharides, molecular ratios and molecular weights are: Le-2-1, Glc: Gal: Man: Xyl: Ara: Rha = 86.05: 2M: 2.06: 1.96:1.98: 1.58 MW = 5.7 x 1O3; Le-2-2, Glc: Gal: Man: Xyl: Ara: Rha = 70.60: 10.25: 8.51: 1.80: 2.38: 3.16 MW = 5.8 x 1O4.
