Abstract: Lentinula edodes (Berk) Pegler showed maximum radial growth at 25°C on pH 5. Among the carbon and nitrogen sources evaluated, optimum mycelial growth was obtained on starch and urea. Spawn was prepared on sawdust of Delbergia sisso, Acacia and Populas spp. Of these saw dust of Delberia sisso proved most suitable. Three types of saw dust were blended with wheat chaff, rice straw and cotton waste for spawn running. All saw dusts amended with cotton waste were found to be superior than wheat chaff and rice straw amendment. The initial protein of substrate ranged between 0.94 to 3.75% which increased with spawn running due to impregnation of substrate with fungal mycelium.
