Abstract: Isozyme analysis is an usefull tool for genetic studies related to agronomical research. A method lying on isoelectrofocusing followed by a blotting onto nitrocellulose was improved in order to increase the sensitivity and the reproducibility of the enzyme detection. Nine enzyme activities (acetate-esterase, butyrateesterase, TRE-phenoloxidase, DRE-phenoloxidase, peroxidase, alcohol dehydrogenase, malate dehydrogenase, diaphorase and acid phosphatase) were studied on some industrial strains of Agaricus and Pleurotus species. The results allowed the determination of the polymorphism rate of each pair activity / species and the selection of the most suitable enzyme activities in order to characterize the strains belonging to each species studied. The enzyme polymorphism pointed out allowed the unambiguous characterization of each dikaryotic strain smdied, even for closely related strains. "Identity cards" specific of each strain were established in order to allow, at longer range, their industrial protection.
