Abstract: The mitochondria of the edible basidiomycete Agrocybe aegerita were purified by differential centrifugations followed by a density gradient fractionation. A mitochondrial (mt) DNA purification procedure was developped from isolated mitochondria of a wild-type dikaryotic A. aegerita strain. A representative library of this mtDNA molecule was realized in E. coli by using the plasmid vector pBR322. Recombinant DNA molecules between vector and mtDNA Hindin restriction fragments were analysed and characterized by agarose gel elctrophoresis and Southern hybridizations onto nitrocellulose sheets, with the 32P-labeled-mtDNA molecule. Summation of the cloned restriction fragment sizes gave a mitochondrial genome size equal to 76,000 ± 3,000 bp.
