Abstract: Mitochondrial genotypes were examined in 29 commercial strains of Agaricus bisporus. Only three genotypes were found and the majority of the strains shared one mt genotype indicating the low genetic variability among commercial strains. To study the contribution of the mt genome to the vegetative and generative fase of cultures, mitochondrial inheritance was studied in heterokaryons obtained by mating and protoplast fusion. The results showed that in A. bisporus, mitochondria are uniparentally inherited. The same mt genotype was consistently inherited in matings. However, in the same heterokaryons obtained after fusion of protoplasts, either one of the mt genotypes was recovered. In two cases, recombinant mt DNAs were recovered. Possible mechanisms of mitochondrial inheritance in A. bisporus are discussed.
