BIOTECHNOLOGICAL DIRECTIONS IN POLISH BREEDING OF RUBUS

Abstract:
Two molecular techniques, RAPD and ISSR, were used to analyze genetic relatedness among thirty-six Rubus cultivars and clones in order to determine their suitability for breeding. All of the genotypes could be identified and distinguished from each other using eleven of the 60 RAPD and 16 ISSR primers tested. The dendrograms generated on the base of RAPD and ISSR data showed generally similar tendency, although there were sometimes significant differences in the percentages of relatedness obtained. The genotypes belonged to several clusters (black raspberry, Hungarian cultivars from ‘Fertodi’ group, selected Polish crosses, ‘Algonquinn’ and ‘Latham’, and subgroup of raspberry and blackberry hybrids). An embryo rescue technique was used to regenerate plants from interspecific crosses within Rubus genus. The experiment was conducted on embryos from eight cross combinations between distantly related Rubus species. Finally, plants with fully developed organs were obtained from 29% of isolated embryos. The efficiency of the process for control crosses was 66%.
