DEVELOPMENT OF PRIMARY CORE COLLECTION FOR DIOSPYROS KAKI THUNB. BY STEPWISE CLUSTERING

Abstract:
China preserves many accessions of persimmon, Diospyros kaki Thunb. The large amount of accessions has hindered the effective management, evaluation and utilization of this available germplasm. The objective of this study was to develop a primary core collection for persimmon to assist the breeding program. The Chinese National Persimmon Germplasm Repository is located on a farm affiliated with the Northwest Sci-tech University of Agriculture and Forestry, Yangling, Shaanxi Province. From this genebank, 558 accessions were used as experimental materials in our investigation. The genotypes were separated into groups based on astringency and geographical distribution. The pollination constant non-astringent (PCNA) type was screened into the primary core collection directly. For the 4 subgroups of pollination constant astringent (PCA) type, stepwise clustering was performed by unweighted pair group method with arithmetic mean (UPGMA). Then, the primary core collection, comprised of 180 accessions, was developed and evaluated. Five parameters, the mean, range, variance, coefficient of variation and χ2 value, were used to validate representatives of the primary core collection. The results showed that 180 accessions could represent the entire collection of persimmons. The primary core collection will be useful in future breeding efforts.
