DISEASE-FREE PRODUCTION AND MINIMAL-GROWTH PRESERVATION OF IN VITRO BANANA (MUSA SPP.)

Abstract:
The aim of this research was to investigate the disease-free production and in-vitro preservation of banana. Shoots of banana plantlets were cultured on MS media supplemented with 0.288 µM gibberellic acid (GA) or without GA and then incubated at 36°C in the dark. The results showed that the shoot of plantlets cultured on GA-treatment were 1.48 times longer than those cultured on medium without GA. The meristem-tips (0.5 mm in size) were then dissected and cultured on slant modified-MS media containing 8.88 µM benzyl aminopurine (BA) and 150 ml coconut water. The meristem-tips were completely free of contamination and 17.5% recovered growth on modified-MS media after 14 days. The disease-free plantlets derived from meristem culture were used as initial plant material for in vitro preservation. The clumps of disease-free plantlets were cultured on MS media supplemented with 0, 34, 68 or 136 µM paclobutrazol (PBZ) cultured at 10 or 25µC as a minimal growth treatment for long-term preservation. In vitro banana preservation at 10°C were used to slow growth and development. Shoot height of plantlets grown at 10°C was significantly lower (2-6 times) than those plantlets cultured at 25°C. At 25°C, shoot height, root length and leaf area of plantlets were negatively correlated to PBZ concentration in the culture media, while chlorophyll concentration was positively correlated. Suggested treatments for banana preservation were either at 10°C or at 25°C with the addition of 136 µM paclobutrazol.
